New Solutions for 16S Metagenomic Sequencing

Depletion of Human DNA and Reduction of Bacterial Contamination towards a Standardised Method for Infection Diagnosis with Next Generation Sequencing (Edelmann et al. 2018)

Edelmann and colleagues compared 4 DNA extraction kits and the influence of human DNA removal for use in low microbial load microbiome analysis. Additionally, two master mixes were tested for bacterial DNA contamination.

The authors found out that DNA extraction and master mix reagents have a huge impact on the contamination signature in sequencing reads. They resumed that MolYsis-SelectNA™plus together with NGS Mastermix 16S V3/V4 aid an efficient workflow for NGS towards a standardised method in microbiome infection diagnosis. An efficient workflow towards a standardised use of NGS in infection diagnosis was proposed.

Amplicon PCR for 16S library preparation

The ultra-clean NGSeq 16S V3/V4 is a master mix for the amplification of eubacterial DNA with primers specific for the V3/V4 region of the 16S rRNA gene. Primers also include overhang adaptor sequences suitable for the library preparation for 16S metagenomic sequencing with Illumina® MiSeq system. The NGSeq 16S V3/V4 includes everything necessary to quickly perform the Amplicon PCR as first stage of the 16S library preparation protocol.
